Roche NimbleGen Instruments Roche NimbleGen now presents a complete, optimized workflow for NimbleGen DNA microarrays. This optimized workflow encompasses high-density NimbleGen arrays, 4-Bay and 12-Bay Hybridization Systems, Microarray Dryer, MS 200 Microarray Scanner, NimbleScan and SignalMap software , and a comprehensive range of kits and consumables to facilitate processing of NimbleGen DNA microarrays from hybridization through data analysis. The NimbleGen 4-Bay and 12-Bay Hybridization Systems are designed for highly reproducible hybridization conditions with precision controlled temperature and active mixing to provide high sensitivity and specificity hybridization results. The Mixers, a line of adhesive sample enclosures that provide containment and active mixing during hybridization, increasing both signal and stringency, are available in a range of configurations to support single sample (2.1M and 385K) and multiwell (3x720K, 12x135K, and 4x72K) array formats. The NimbleGen Microarray Dryer helps to ensure production of high-quality images with minimal residue by completely removing wash solutions prior to scanning. For NimbleGen Sequence Capture arrays, the NimbleGen Elution System facilitates the elution of enriched DNA from the surface of the following hybridization and washing procedures. The MS 200, a high-sensitivity laser scanner, reproducibly acquires two-color fluorescent image data down to 2 micron pixel resolution and allows completely automatic high-throughput image processing. Effective washing and complete removal of wash solution prior to scanning is critically important when processing DNA microarrays. Salt crystallization and other residue formations resulting from insufficient washing or removal of wash solution can compromise vital data, contribute to high background signal, or both. Additionally, protection of the hybridized array from environmental degradation during the wet-to-dry transition is essential to maintain fluorescent signal. The NimbleGen Microarray Dryer helps to ensure production of high-quality images with minimal residue without the use of noxious chemicals, such as acetonitrile. The array drying step is performed in a protected environment to reduce atmospheric and light-induced degradation of Cy dyes. A key component of the NimbleGen microarray workflow, the NimbleGen Microarray Dryer is used with NimbleGen Hybridization Systems, Mixers, and Wash Buffer Kits to provide the complete array processing solution for NimbleGen DNA Microarrays. The NimbleGen Elution System (shown at right) is used in conjunction with an Elution Chamber (provided separately with each NimbleGen Sequence Capture array) to facilitate the elution of enriched DNA from the surface of a NimbleGen Sequence Capture array following hybridization and washing procedures. Please refer to the NimbleGen Arrays User’s Guide: Sequence Capture Array Delivery (Version 2.0) for more details). The Elution System Retaining Ring secures the Elution Chamber and array firmly against the surface of the Elution System Base, while the Elution System Base conducts heat to the array surface to release captured DNA from the oligonucleotide probes and into solution.
